Can someone explain the difference between these two?

I should know this, but how are these played differently?  The first is obvious, but do you skip the

G string on the second?

2 ways : 

 

Either you do finger picking with the thumb and index, if the song is meant to be played like that.

 

More commonly with a pick , you mute the middle string with the finger on the 3 lying on it without pressing it to the fret.

It can also be represented like this

Rocksmith tries to recognize the muted string then, and some users believe it often fails.

 

To bypass this apparent problem, some use

So Rocksmith only tries to recognize 6 and 3, and cares less for the muted string.

But it has to be muted.
