What version of Windows? If Win 7 or 8 you can try adding an exception in DEP (Data Execution Prevention) for Rocksmith2014.exe I won't go into how to do it here, it's very easy to do http://windows.microsoft.com/en-us/windows7/Change-Data-Execution-Prevention-settings?SignedIn=1

Otherwise check in Programs and Features (select show updates) for recently installed Windows updates. Two posters that are reporting the issue attempted to run after Tuesday (Windows Update day), I am assuming this was first run since an update. If updates were installed after last known working run of Rocksmith I'd uninstall those updates and attempt to run Rocksmith again, if it works go into Windows updates and begin to manually install updates one to three at a time testing Rocksmith after each update install. This will allow you to isolate which update is interfering. This of course assumes it is a Windows Update issue.

If it were me, I'd try DEP first if it works you can leave it, your system will work just fine though I would still need to track down the reason for the sudden change.
